Thioglycolate medium is prepared according to the formulation indicated by the harmonized method EP, USP, JP for the sterility test of pharmaceutical products but without resazurine. Cystine and sodium thioglycolate, at a concentration in which they have a reduced toxicity for microorganisms, act as reducing substances reacting with the oxygen molecules from the soil and eliminating and preventing the accumulation of peroxides, lethal for some aerobic and anaerobic microorganisms. The sulfhydryl groups (SH) of the two substances also inactivate the mercurial compounds, the compounds based on arsenic and other heavy metals, integrated in a low redox potential and ensuring anaerobic conditions. The agar present at a concentration of 0.75%, helps in the initialization of the growth of the anaerobes and provides their growth starting from low inoculations; it also delays the diffusion of oxygen in the medium and serves only a free release from the soil of the reducing substances produced by microorganisms and carbon dioxide. Casein peptone and yeast extract are sources of nitrogen, carbon, vitamins and minerals for microbial growth, glucose is a source of carbon and energy, sodium chloride is provided by an adequate osmotic content in the broth of culture. The soil can be integrated with various compounds, including Vitamin K to stimulate the growth of some strains of Bacterioides and Prevotella and hemin, as a source of factor X, to increase microbial growth.
