Pseudomonas aeruginosa are gram-negative micro-organisms that grow on selective media containing cetrimide and produce pyocyanin, or micro-organisms that grow on selective media containing cetrimide, are oxidase positive, fluoresce under UV light (360 ± 20) nm, and are able to produce ammonia from acetamide. Colonies producing pyocyanin on CN Agar medium are considered as Pseudomonas aeruginosa. The other colonies producing fluorescence or other reddish-brown pigmented colonies have to be confirmed on the King B medium, for the demonstrating of the synthesis of a specific pigment: the pyoverdin. King B medium contains high strength of magnesium sulphate which provides the cations necessary for activation of pyoverdin, which gives the culture medium fluorescent greenish yellow colour. Most strains (98%) produce a water-soluble fluorescent pigment. Thus, any colony appearing on the plate within 5 days in the presence of any fluorescence shall be recorded as positive. The base medium is in powdered format to be supplemented
