Kovacs reagent is intended for the demonstration of indole production by bacteria possessing a tryptophanase (Gram negative bacteria, especially Enterobacteriaceae). Bacteria possessing a tryptophanase are able to breakdown tryptophan into indole. When reacting with this indole, Kovacs reagent turns a cherry red colour. The reagent must be added to a bacterial culture started from a colony isolate grown in Peptone Water indole free or Tryptophan broth.
