BCYE is a non-selective medium for the confirmation of Legionella spp. and Legionella pneumophila in water (water intended for human consumption, hot water for sanitation purposes, water for industrial use, natural mineral waters for thermal purposes, water for recreational activities, etc.). Legionellae are gram-negative bacteria requiring L-cysteine for growth. The principle of the medium relies on the ability of Legionella to multiply on buffered agar containing yeast extract, activated charcoal, L-cysteine, alpha-ketoglutarate and iron. Legionella form grey-blue colonies varying sometimes to yellow, green, white, brown, purple and pink. Bio-Rad BCYE is available in ready-to-use 90 mm plates with or without L-cysteine and also as 90 mm biplates: Legionella grow only on L-cysteine supplemented half of the BCYE biplate dish or BCYE with L-cysteine.
